MCP-ADAR2dd(E488Q)

MCP-ADAR2dd(E488Q) is a fusion protein consisting of the MS2 bacteriophage major coar protein (MCP) linked to the deaminase domain of ADAR2(E488Q) via a Gly-Ser linker. It is used as a component of the DART VADAR circuit [1].

The MCP domain binds MS2 mRNA hairpins and recruits the deaminase domain to its mRNA target. The ADAR2dd domain deaminates mismatching adenine bases in mRNA duplexes to an inosine base. This principle is used by the DART VADAR technology to create mRNA sensors [1] by editing a UAG stop-codon to a UIG codon, which is read as a sense UGG codon by ribosomes.
